SCL Expression in the Mouse Embryo Detected With a Targeted
نویسندگان
چکیده
The helix-loop-helix transcription factor SCL (TAL1) is indispensable for blood cell formation in the mouse embryo. We have explored the localization and developmental potential of cells fated to express SCL during murine development using SCL-lacZ mutant mice in which the Escherichia coli lacZ reporter gene was ‘knocked in’ to the SCL locus. In addition to the hematopoietic defect associated with SCL deficiency, the yolk sac blood vessels in SCLlacZ/lacZ embryos formed an abnormal primary vascular plexus, which failed to undergo subsequent remodeling and formation of large branching vessels. Intraembryonic vasculogenesis in precirculation SCLlacZ/lacZ embryos appeared normal but, in embryos older than embryonic day (E) 8.5 to E9, absolute anemia leading to severe hypoxia precluded an accurate assessment of further vascular development. In heterozygous SCLlacZ/w embryos, lacZ was expressed in the central nervous system, vascular endothelia, and primitive and definitive hematopoietic cells in the blood, aortic wall, and fetal liver. Culture of fetal liver cells sorted for high and low levels of b galactosidase activity from SCLlacZ/w heterozygous embryos indicated that there was a correlation between the level of SCL expression and the frequency of hematopoietic progenitor cells. r 1999 by The American Society of Hematology.

Targeted gene disruption experiments in the mouse have demonstrated an absolute requirement for several transcription factors for the development of hematopoietic progenitors during embryogenesis. Disruption of the basic helix-loop-helix gene SCL (stem cell leukemia) causes a block early in the hematopoietic program with defects in all hematopoietic lineages. To understand how SCL participates ...
متن کاملThe effects of electromagnetic fields on alpha-fetoprotein expression in the amniotic fluid of mouse embryo
Amniotic fluid (AF) is essential for fetal development and maturation during pregnancy. The levels of proteins in AF have been determined in many studies to screen for potential biomarkers of pregnancy-associated abnormalities. Alpha-fetoprotein (AFP) is a major AF and plasma protein produced by the yolk sac and the liver during the fetal period. APF serum concentrations are commonly used for s...
متن کاملP-67: Quantitative Expression of Pluripotency Specific-Genes in Mouse Blastocysts Produced by In Vitro Fertilization
Background: The efficiency of in vitro fertilization (IVF) is still low to be developed to blastocyst stage probably because of environmental conditions. It is likely that in vitro environment can not exactly mimic in vivo environment due to differences in media, metabolic content, atmospheric composition, temperature and pH. Therefore it may affect embryo quality by changing in embryo gene exp...
متن کاملP-117: Gene Expression and Developmental State of Mouse Cloned Embryos after Treatment with Histone Deacetylase Inhibitor,Suberoylanilide Hydroxamic Acid (SAHA)
Background: It is known that acetylation level of the nuclear histones in cloned embryos is lower compare to normally developed embryos. Histone deacetylas inhibitors (HDACi) with improvement of acetylation level in these embryos can affect embryo quality in pre-implantation stage and expression level of different genes especially developmental genes. Materials and Methods: In this research, SA...
